Triple Resonance Backbone Assignment

The description here assumes that the backbone assignment will be carried out using CBCA(CO)NH and CBCANH spectra. Many of the steps are the same if using HNCA, HN(CO)CA, HNCO and HN(CA)CO spectra, and differences are highlighted in a separate section.

Now comes the actual assignment part which follows the process outlined in the theory pages. First you are shown how to do the assignment manually by going through through each step one at a time. Once you feel comfortable with the general process you can speed things up by using the semi-automated Protein Sequence Assignment function. This simply condenses several steps into one, thus speeding the process up.
